Cold-chain storage best practices for research peptides

Temperature, light and humidity are the three quiet enemies of peptide stability. A short field guide to handling, transport and storage of research peptides in a laboratory setting.

Once a research peptide has been synthesised, purified, lyophilised and analysed, the stability of the finished material is entirely a question of how it is stored. Three environmental variables matter more than anything else: temperature, light and humidity.

Temperature

Most research peptides are stable for years in their lyophilised form when kept at −20 °C. For active laboratory use, refrigeration at 2–8 °C is the appropriate operating temperature for unopened vials and is the condition under which Spheres Peptides ships its catalogue. Avoid repeated freeze-thaw cycles of reconstituted solutions, as each cycle introduces an opportunity for aggregation and adsorption losses.

Light

UV and short-wavelength visible light can drive photo-oxidation of aromatic residues — tryptophan and tyrosine in particular — and accelerate degradation of redox-sensitive groups. Storing vials in their original carton, or in any opaque container, eliminates this pathway. Where vials must be kept on bench during a working day, an amber glass beaker or aluminium foil sleeve is sufficient.

Humidity

Lyophilised cakes are hygroscopic. Even brief exposure to humid laboratory air will draw moisture into the powder, raising water activity and accelerating hydrolysis of labile bonds. The practical implication is that vials should remain capped until the moment of reconstitution, and any partial-use reconstitution should be planned to consume the cake in a single session rather than re-sealing and re-opening over several days.

Transport and receiving

When a peptide shipment is received it should be unpacked promptly, the vials inspected for transit damage, and any vials with melted ice packs, broken seals or visibly collapsed cakes flagged before being placed into stock. A short stock-handling checklist

  • Log every received batch into your laboratory inventory with the batch number, COA reference and arrival date.
  • Store unopened vials at 2–8 °C, protected from light, in the original carton.
  • Keep a written record of the date each vial is opened and reconstituted.
  • Discard reconstituted solutions in accordance with the laboratory protocol governing that work.
  • Treat any vial with a damaged seal, discoloured cake or unexplained loss of vacuum as compromised.
